The Defense Advanced Research Projects Agency (DARPA) is soliciting proposals to develop an in-silico capability for mapping all protein-protein interactions. Utility of this capability will be demonstrated in identifying novel diagnostic and therapeutic targets within the interaction space underlying including traumatic brain injury.
Performers will develop an in-silico capability to map protein-protein interactions (PPIs) by creating a low-dimensional 'diffusive binding landscape' signature. In Phase 1, performers will capture an initial interactome for TBI-associated proteins, modeling ~100,000 PPIs over >10 conditions. In Phase 2, performers will scale to a PPI hypercatalog of 10 million unique interactions modeled over >50 unique cellular contexts, capturing stable dimers, transient interactions, multiprotein complexes, and biocondensate/domain formation. The work includes internal benchmarking, validation against government team datasets, and iterative refinement of PPI predictions.
