DNA sequencing of commercially important groundfish for the National Marine Fisheries Service (NMFS) Alaska Fisheries Science Center (AFSC).
The contractor shall perform laboratory processing (DNA extraction, library preparation) and providing raw sequence data of DNA samples and long-read sequencing for 45 Pacific cod samples and 6 long-read samples. Short-read sequencing must be on Illumina NovaSeq X Plus or equivalent to 150bp PE150 at 8-15x depth per sample. Long-read sequencing must be on PacBio Revio or Oxford Nanopore accumulating to a minimum of 20x coverage per fish and 13Gb per sample with median consensus accuracy ≥Q30. Deliverables include FASTQ files, sample metadata, and quality control reports.
